The operation was a response to an increase in local prostitution services being offered over the Internet, police said. They declined to release the name of the hotel.
Investigators worked in cooperation with hotel staff, securing two rooms and posing as customers, police said.
The two women arrested were Ludivina Ponce, 27, of Madera, and Jillian Kayleigh Ruff, 22, of Sacramento, police said.
They were later released on their own recognizance pending court appearances.
